Potencialidades de las levaduras en el aprovechamiento de los residuales del café
Resumen
Se realiza una caracterización de los residuales del beneficio húmedo del café atendiendo a su posible aprovechamiento mediante levaduras, encontrándose que con las tecnologías tradicionales sólo es factible de utilizar la pulpa de café. Igualmente se encontró que el proceso tecnológico y el tiempo de fermentación influyen sobre su composición. De estos residuales se aisla una cepa de Saccharomyces cerevisiae con actividad pectinolítica. La síntesis de enzimas pécticas por esta cepa está asociada al crecimiento y es estimulada por la presencia de pectina en el medio de cultivo desde 0,1- 10g/L, no observándose diferencias significativas en las producciones para las diferentes concentraciones de pectina en el medio, excepto para pectina 5 g/L donde la actividad enzimática resultó significativamente superior.
La enzima péctica de esta cepa fue purificada 40,2 veces por cromatografía de intercambio iónico en Fractogel EMD Sulfonato- 650 (s) mostrando homogeneidad electroforética en PAGE-SDS y una masa molecular de aproximadamente 60 kDa. Se obtuvieron los valores de Km y Vm, siendo estos 1,9mg/mL y 312,1 nmol/mL.min para la pectina (28,9 % de esterificación) y 0,7 mg/mL y 205,1 nmol/mL.min para el ácido poligalacturónico. La enzima se comporta como una poligalacturonasa tipo endo y presenta un rango de pH y temperatura óptimos de 4-4,5 y 55 0C respectivamente. A pH 3 se conserva más del 80 % de la actividad y a 37 0C se mantiene estable durante 60 minutos, inactivándose completamente a pHs superiores a 6,5 y a 60 0C en 10 minutos. La producción de alcohol en medio convencional por esta cepa muestra una eficiencia superior al 86 %, no influyendo significativamente sobre ésta la presencia de pectina en el medio a una concentración de 10 g/L. La cepa sintetiza activamente pectinasas durante la fermentación alcohólica e hidroliza extensamente la pectina del medio sin consumo aparente de los productos de hidrólisis. La cepa es capaz de degradar la cafeína en más de un 85 % en un medio mineral cuando la utiliza como única fuente de nitrógeno, y puede utilizar la pectina en presencia de glucosa 5 g/L. The production of alcohol in conventional medium by this strain shows an efficiency greater than 86 %, the presence of pectin in the medium at a concentration of 10 g/L not significantly influencing it. The strain actively synthesizes pectinases during alcoholic fermentation and extensively hydrolyzes pectin from the medium without apparent consumption of the hydrolysis products. The strain is capable of degrading caffeine by more than 85% in a mineral medium when it is used as the only nitrogen source, and it can use pectin in the presence of 5 g/L glucose.
